Carrier free antibodies

Antibodies are offered in a carrier free solution from Cell Signaling Technology (CST) to be used in multiplexed single cell experiments and imaging (mass cytometry applications). The antibodies are:

• Validated in relevant applicaions such as flow and/or IHC
• Validated for specificity, sensitivity and reproducibility
• Free of BSA and other carrier proteins that can interfer with heavy metal conjugation
• Available in high concentrations (>1mg/ml), which is minimum requirement in order to conjugate the antibody​​

Carrier free antibodies

When choosing antibodies for MaxPar® metal conjuation in applications such as CyTOF, single cell, Helios™ (Fluidigm) or imaging Hyperion™ (Fluidigm), it is important that the antibodies already are validated in relevant applications. Antibodies from CST are subject to strict validation and specificity testing in order to ensure reproducible results you can trust. The antibody you choose should be matched to your sample preparation method. For FFPE samples you can select from CST`s portfolio of IF-paraffin or IHC-validated antibodies, while for frozen tissue you can consider the IF-Frozen validated antibodies.

ALK and ROS1

Anaplastic lymphoma kinase (ALK) and c-ros oncogene 1 (ROS1) are two proteins of the same family of receptor tyrosine kinases.

Research studies have shown that mutated versions of ALK and ROS1 proteins can function as oncogenic drivers, and that these proteins are expressed in various forms of cancer, as for example non-small cell lung cancer (NSCLC).

Cell Signaling Technology (CST) offer two very sensitive antibodies for detection of ROS1 and ALK proteins. The antibodies are produced by immunizing animals with C-terminal epitopes, which makes it possible to detect both full-length proteins as well as the mutated C-terminal fusion proteins. The antibody sensitivity is so high that you may detect endogenous levels of ROS1 and ALK. The antibodies does not cross-react with other related proteins when analyzed by western blot.

SSX and SS18-SSX

SS18 is a protein that has been shown to be a part of the SWI/SNF complex. The SS18-SSX fusion proteins are a result of in-frame fusions that fuse the SS18 gene on chromosome 18 with X chromosome genes SSX1, SSX2, and to a lesser extent SSX4.

Human synovial sarcoma (SS) accounts for 8 - 10 % of all soft tissue malignancies and 95% of these malignancies express the recurrent translocation of the SS18 gene on chromosome 18.

Cell Signaling Technology (CST) offers an antibody for specific detection of the fusion protein: SS18-SSX (E9X9V) XP® Rabbit mAb (#72364). This antibody recognizes endogenous levels of the SS18-SSX fusion protein. This antibody does not cross-react with wild-type SS18 or SSX proteins.

CST also offer a SSX (E5A2C) Rabbit mAb (Carboxy-terminal Antigen) antibody (#23855) recognizes endogenous levels of total SSX1-4 proteins. This antibody also detects the SS18-SSX fusion protein.
