The Nucleofector™ technology uses a specific combination of electrical parameters and solutions that are optimized and specific for different cell types. This cell-specific combination enables transfer of a molecule directly into the cells’ nucleus.
Thus, this type of transfection is independency from cell proliferation, and this allows for efficient transfection of even non-dividing primary cells (like resting T cells or neurons) and also speeds up expression.
The phosphate backbone of nucleic acids carries one negative charge per residue. As a result, nucleic acids are highly negatively charged. The chemicals in transfection reagents (polymers or lipids) coats the DNA and forms a DNA:transfection reagent complex or nanoparticles, which protect nucleic acids from nuclease-mediated degradation. This process makes it easier for the DNA:transfection reagent complex to cross the cell membrane, and be released into the cytoplasm or nucleus.
