Since 1985, as a recognized oligo synthesis company, Eurogentec have been producing a wide range of custom Taqman® Probes. Their extensive portfolio of fluorescent dyes and NFQ quenchers allows our dual-labeled probes to be utilized across any qPCR channels, from deep blue to infrared. In addition to classical fluorophores and quenchers like HEX, FAM, TAMRA, and BHQ®, they offer custom qPCR probes labeled with our proprietary dyes (HiLyte™ Fluor, QXL®) using an IP-friendly approach for commercialization.

Eurogentec offer classical and locked nucleic acid Taqman® probes for real-time qPCR. A variety of fluorescent dyes are available depending on the selected quencher, such as:

MGB-probe principle

MGB Probes have a fluorescent reporter dye and a quencher at their 5’ and 3’ ends, respectively.

During the amplification process, the 5’>3’ exonuclease activity of the Taq DNA polymerase cleaves the fluorophore from the probe. Since the fluorophore is no longer subjected to FRET quenching, it starts to fluoresce. This fluorescence can be measured, and the level is directly proportional to the amount of target DNA accumulating during the PCR reaction.

Molecular Beacons

As a licensed supplier of Molecular Beacons, Eurogentec offer standard, wavelength-shifting, and 2' O-Me RNA Molecular Beacon probes. Their custom-synthesized Molecular Beacons are available with DABCYL or BHQ® non-fluorescent quenchers, with additional dye options available upon request.

Molecular beacons principle

Molecular Beacons are specialized probes featuring a stem-loop structure with a fluorophore at the 5’ end and a quencher at the 3’ end. The stem sequence holds the fluorophore and quencher in close proximity, preventing fluorescence in the absence of a complementary sequence to the loop region. In this state, any photons emitted by the fluorophore are absorbed by the quencher.

When a complementary target sequence is present, the probe unfolds and hybridizes with the target. This separates the fluorophore from the quencher, allowing the fluorophore to emit light. The resulting fluorescence is directly proportional to the amount of target sequence present and is measured in real-time, enabling the quantification of the target sequence.
