Q5® high-fidelity DNA polymerase sets a new standard for both fidelity and robust performance.

    Achieve precision without sacrificing speed. Q5's unique design incorporating the SSo7d processivity-enhancing domain enables shorter extension times, as low as 10 seconds per kb. Additionally, aptamer-based hot start requires no initial denaturation step and enables room temperature setup.
  • Amplify templates up to 20 kb
    With Q5, you can reliably amplify simple templates up to 20 kb. Complex templates up to 10 kb can also be amplified with a high degree of confidence.

NEBNext® Ultra™ II Q5® Master Mix (#M0544)

NEBNext Ultra II Q5® Master Mix is the most recent NEBNext formulation of Q5 High Fidelity DNA Polymerase, optimized for amplification of NGS libraries. This hot start formulation is designed for robust, high-fidelity amplification of next-generation sequencing (NGS) libraries, and further improves the uniformity of amplification of libraries, including superior performance with GC-rich regions.

NEBNext Ultra II Q5 Master Mix is also included as a component in all NEBNext Ultra II kits, for DNA and RNA library preparation.

NEBNext® Q5U® Master Mix (#M0597)

NEBNext® Q5U® Master Mix contains a modified version of Q5® High Fidelity DNA Polymerase, optimized for amplification of uracil-containing NGS libraries. This hot start formulation is designed to read through uracils, providing robust, high-fidelity and uniform library amplification. 
NEBNext Q5U Master Mix is included as a component in the NEBNext Enzymatic Methyl-seq Kit (NEB #E7120) and is also compatible with library amplification in bisulfite sequencing workflows.

Q5U® Hot Start High-Fidelity DNA Polymerase (#M0515)

Q5U Hot Start High-Fidelity DNA Polymerase is a modified version of Q5® High-Fidelity DNA Polymerase, a novel thermostable DNA polymerase that possesses 3′ to 5′ exonuclease activity, and is fused to a processivity-enhancing Sso7d domain. Q5U contains a mutation in the uracil-binding pocket that enables the ability to read and amplify templates containing uracil and inosine bases.

  • Achieve superior amplification of bisulfite-converted, deaminated, or damaged DNA (e.g., FFPE)
  • Prevent carryover contamination when combined with dUTP (#N0459) and Uracil DNA Glycosylase (UDG) (#M0372) treatment
